Function

ANO6 (TMEM16F), a calcium-activated phospholipid scramblase and ion channel of the anoctamin family, localizes to the primary cilium of renal tubular epithelial cells: antibody staining colocalizes with acetylated tubulin in MDCK cells, human collecting duct cells and primary human tubular cells, and the signal is lost after ANO6 knockdown. ANO6 knockdown (about 80%) does not prevent cilia formation or in vitro cyst expansion, but impairs cyst lumen formation of MDCK cells in three-dimensional culture by reducing apoptosis of cells in the center of early aggregates. ANO6 is highly expressed in apoptotic cyst epithelial cells of human ADPKD kidneys (Forschbach et al., 2015, Cell Death Dis, PMID 26448322).
