  • Protein processing & maturation
  • Cilia length regulation

Function

Positively and negatively controls both ciliary length and the activity of the Hh pathway, regulates a broad array of processes at levels ranging from the subcellular to the organismal (28817564). Length of wild-type and Ccrk mutant cilia under steady-state (serum starved) conditions measured, although the average cilia lengths were generally similar (2.6 and 3.5 μm, for control and mutant cilia, respectively), there was a broader distribution of cilia lengths in the mutant cells than in the control cells (std. dev. of 0.78 vs. 1.9 for control and mutant cilia, respectively)(PMID: 28817564).

Model organism evidence

C. elegans (3 references)

The conserved CCRK, RCK, and CDKL5 kinases regulate cilia length in diverse organisms.

Mutations in the Domain of Unknown Function 3719 (DUF3719)-containing protein FAM149B1 have recently been shown to elongate cilia via unknown mechanisms and result in the ciliopathy Joubert syndrome.11 Here we identify XBX-4, a DUF3719-containing protein related to human FAM149B1, as a regulator

BROMI/TBC1D32 together with CCRK/CDK20 and FAM149B1/JBTS36 contributes to intraflagellar transport turnaround involving ICK/CILK1.
