Function

FMNL2 is a formin that nucleates and polymerizes actin filaments. A hyperactivating splice mutation in mouse Fmnl2 (nmf418, exon 10 skipping in the autoinhibitory DID domain) causes hydrocephalus, enlarged midbrain colliculi, cerebellar hypoplasia and vertebral defects, whereas Fmnl2 knockout mice are normal. The authors propose that hyperactive FMNL2 disrupts the apical actin network around ependymal basal bodies and ciliary function; this was not tested directly (Zaepfel et al., 2026, bioRxiv, doi 10.64898/2026.10.16.759550).
