Cilia effects upon perturbation of NTRK2

Ciliogenesis screen results (4 screens)

  • Kim2016: No effect
  • Wheway et al. 2015 (siRNA) [siRNA]: Ciliogenesis Defect (z=-2.86) PMID:26167766
  • Breslow et al. 2018 (CRISPR) [CRISPR]: No Significant Effect PMID:29459680
  • Roosing et al. 2015 (siRNA) [siRNA]: Hyper-ciliogenesis (robust z=2.69, ciliated=58.3%) PMID:26595381

Phenotypes

Mouse phenotype:
abnormal rib morphology, decreased prepulse inhibition, increased lean body mass, preweaning lethality, incomplete penetrance, increased circulating hdl cholesterol level
Human ciliopathy phenotype:
obesity, hyperphagia, and developmental delay

Subcellular localization

basal body, cilia, cytosol

Functional category

  • Ciliary assembly/disassembly
  • Trafficking (BBSome, small GTPases, vesicular transport, ATPases)
  • Neurogenesis & migration
  • Signaling (Hedgehog, GPCRs, ion channels)

Function

TrkB localized to the axonemes or basal bodies of cilia only in the presence of BDNF. Axonemal localization, specifically, was abrogated with loss of BBS4. Loss of the ciliary axoneme through depletion of KIF3A impedes activation of TrkB (24867303).
