  • Signaling (Hedgehog, GPCRs, ion channels)

Function

Type II protein arginine N-methyltransferase that monomethylates arginine residues on histones and other substrates; involved in peptidyl-arginine methylation, spliceosomal snRNP assembly, neuronal differentiation, male germline imprinting, and Wnt-pathway regulation, and localizes to cytosol and nucleoplasm (RefSeq). Biallelic variants cause short stature, brachydactyly, intellectual developmental disability and seizures (SBIDDS; OMIM 617157). Per Jiang et al. (2024, Research Square rs-5410799, Fig 6), TTC26 traffics PRMT7 into the primary cilium, where it methylates the Hedgehog transcription factor GLI2; FLNB then imports methylated GLI2 to the nucleus, and this ciliary Hedgehog-GLI2 axis maintains intervertebral-disc extracellular-matrix homeostasis, implicating PRMT7 in adolescent idiopathic scoliosis.
