SENP1 (SUMO-specific protease 1) is identified in this study as a ciliary base-associated deSUMOylase that regulates stress-induced cellular senescence through modulation of FBF1 SUMOylation. The study showed that: endogenous and overexpressed SENP1 strongly localize to the ciliary base in non-senescent cells, super-resolution microscopy demonstrated that SENP1 overlaps with FBF1 at transition fibers, SENP1 physically interacts with FBF1, DNA damage (irradiation) reduces ciliary SENP1 localization and disrupts the SENP1–FBF1 interaction, loss of SENP1 promotes FBF1 SUMOylation and translocation to PML nuclear bodies (PML-NBs), artificial trapping of SENP1 at the ciliary base suppresses stress-induced FBF1 translocation and senescence signaling. Overall, the study identifies SENP1 as a ciliary base-localized regulator of SUMO signaling and senescence pathways(37019904).
