Function

TNF receptor 1, the death receptor for tumour necrosis factor alpha. Wiegering et al. 2026 report the first ciliary localization for this receptor: immunofluorescence of serum-deprived NIH3T3 cells detects TNF-R1 at the basal body and at the daughter centriole, though not within the ciliary axoneme itself. Quantified basal body and daughter centriole signal is unchanged between wild-type and Rpgrip1l-null cells, so the receptor pool is not redistributed when the transition zone is disrupted, which excludes altered death receptor localization as the cause of the raised cell death in RPGRIP1L deficiency. The finding is mechanistically relevant because MOAP1, which is degraded by the proteasome at the ciliary base, is a known interactor of TNF-R1 in the extrinsic apoptotic pathway; the authors propose MOAP1 is recruited to the ciliary base in order to sit close to ciliary death receptors.
