Function

Although nuclear thiol status appears to be unchanged in Tnp mutants, tail structure abnormalities observed in mutant sperm samples indicate an altered thiol status in the flagella. In TPl knockout mutant epididymal sperm, bent midpieces and midpieces with gaps between mitochondria are observed when examined using light microscopy. In TP2 knockout mutant sperm many tails are observed to be unraveled. The same abnormalities are identified at higher magnification using electron microscopy. Observations made during this study also provide evidence that thiol status is altered at some point during cell maturation since structural integrity of mutant sperm midpieces are disrupted following treatment with DTT. After reduction of thiols, mitochondria are noticeably absent from midpieces of TPl knockout mutant sperm. A similar but lesser effect is observed in sperm from the TP2 knockout mutant().
