Function
Profibrotic stimuli disrupt primary cilia on quiescent hepatic stellate cells (HSCs) by inducing XIAP-mediated ubiquitination and proteasomal degradation of IFT88. This contributes to HSC activation and liver fibrosis. Primary cilia exist on multiple cell types in the liver, but only cilia present on HSCs disassemble during liver fibrosis. TGF-β enhances XIAP-mediated ubiquitination of IFT88, promoting its proteasomal degradation. Blocking XIAP-mediated IFT88 degradation ablates TGF-β-induced HSC activation(PMID: 38351372). Endogenous XIAP localizes to primary cilia and the cytoplasm of the ventral forebrain neuroepithelium in E10.5 mouse embryos, and tagged XIAP co-localizes with Ptch1 in primary cilia of NIH3T3 cells; XIAP stays in cilia after Shh stimulation, when Ptch1 leaves, and becomes concentrated at the centrosome during mitosis. XIAP binds the C terminus of Ptch1 in primary cilia and inhibits Ptch1-mediated cell death; XIAP inhibition reduces proliferation, causes cell death and produces a Hedgehog loss-of-function brain and craniofacial phenotype (Aoto and Trainor, 2015, Hum Mol Genet, PMID 25292199).
